Anti Human Globulin Reagent
Blood is vital to man and it is made up of white blood cells, platelets, and red blood cells. Red blood cells are very useful and among other its usefulness is its antigenicity which comes into play in human immune response. Man produces antibodies against antigens he does not possess.
By reason of blood transfusion, pregnancy and other life processes, man is often challenged by immunogenic substances that occasionally give rise to irregular antibodies. Detection and identification of these antibodies are often a great challenge to the transfusion scientist and one of the techniques use in the detection is the use of anti human globulin reagent.
Anti human globulin reagent is a reagent used in the detection of irregular antibodies in human serum. It is a reagent used in blood group serology for performing compatibility testing, antibody screening, antibody detection and detection of Du red cell typing, umbilical card red blood testing.
Anti human globulin reagent is anti-lgG reagent used for the direct anti-globulin test to demonstrate the in-vivo coating of red blood cells with antibody molecules and indirect anti-globulin test to demonstrate the in-vitro coating of red blood cells with antibody molecules as in the detection and identification of unexpected antibodies as well as cross matching tests.
Anti human globulin is a balanced ready to use blend of highly purified immunoglobulins, it contains anti human IgG antibodies and antibodies reactive with human complement C3b and C3d. These anti complement antibodies are IgM class monoclonals and they impart the necessary sensitivity to the reagent.
ANTI HUMAN GLOBULIN: BACKGROUND OF THE STUDY
The use of anti human globulin was first described by Moreschi in 1908 (Moreschi 1908). This reagent is being produced in European counties and it is imported into Nigeria. The reagents pass through different supply chain before we finally get to use it in the various laboratories in our country.
Hence it is proper to embark on local production of reagents in our country.
ANTI HUMAN GLOBULIN: STATEMENT OF THE PROBLEM
More often some reagents e.g AHG may show some error in production by the manufactures or the ill handling and storage by the retailers techniques used by scientist.
Therefore the reason for this project is to know whether the problem arises from the manner of production or otherwise.
AIMS AND OBJECTIVES
- Attempt to prepare anti-human globulin locally.
- To compare locally prepared anti human globulin reagent against commercial ones.
- To improve locally prepared anti-human globulin reagent.
- It will create an avenue to teach others how to prepare anti human globulin reagent locally.
Successful attempts at this project will give scientist the zeal to prepare reagents locally and not to depend on the commercial product. The knowledge gotten from this project will help to reduce unemployment since the practice of local preparation of these reagents can serve as source of income.
It brings out the spirit in us as scientists to embark in research on how to locally prepare other reagents.
