Fish Hatchery Acclimatization And Selection
Breeders sourced brooder stock and fed diet of (30-35% protein at 3.5% body weight. At this point the male and female brood stock can be selected in the morning and stocked separately in smaller holding tanks or pond, because it make catching easy during the actual operation.
Hormone Collection In Fish Hatchery
Different types of hormone are used to induce breeding of catfish, which include;
i. Human chronic gonadotropin (HCG).
ii. Releasing hormone analogue (RHA).
iii. Pituitary gland extract (PGE) from catfish or carp.
iv. Ovaprim (Canada).
v. Ovatide (India)
vi. Ovary-prim (China).
All this hormones are for inducing or achieving a complete ovulation of the female catfish, but hormone dosage depends on the size/ weight of the brood stock used. Hormone that is going to be used should be stored in refrigerator and only the female brood stock is injected.
Timing and injection in fish hatchery
The chosen hormone should be injected to the female brood stock intramuscular, the ideal time for injection is morning or evening and timing is between 8-9 hours intervals depending on the prevailing temperature, since the lower the temperature, the higher the hour for complete ovulation. The actual brood stock (that is the male and female fish) should be one year and above and at appropriate equal body weight for successful fish breeding or hatching.
Gametes Collection / Stripping And Fertilization In Fish Hatchery
After 8-9 hours of hormone injection, the ovulated eggs are stripped manually by gentle application of pressure on the lower abdomen of the injected female fish. The male catfish are sacrificed to get the male gonad from which the milt (male gamete) is collected.
The male gamete collected can be stored in a refrigerator at 40C for 24 hours or used direct by pouring it into the bowel containing the stripped eggs. The ovulated eggs are mixed with milt by gentle shaking or with plastic spoon and water saline water is added to ensure homogenous mixing as stirring is kept continuously.
